耐受性树突状细胞治疗卵清蛋白致敏小鼠的实验研究 被引量:4

Therapeutic effects of tolerogenic dendritic cells on ovalbumin allergic BALB/c mice

摘要 目的 将重组AdCTLA4Ig/Adα4β7诱导的耐受性树突细胞(DC)回输于卵清蛋白(OVA)致敏小鼠体内,观察其对食物过敏小鼠的治疗效果,为临床治疗儿童食物过敏提供新的思路.方法 建立BALB/c小鼠食物过敏模型.BALB/c雌鼠24只,分为3组:肠道激发4 h后回输耐受性DC为治疗组,回输生理盐水为阴性对照组和OVA过敏小鼠为阳性对照组.ELISA测定各组血清中OVA特异性IgE水平;HE染色观察空肠形态;免疫组化法检测小鼠小肠组织中白介素10(IL-10);免疫荧光法检测转化生长因子β(TGF-β)表达水平.结果 ELISA法检测小鼠血清中OVA特异性IgE水平阳性对照组为(0.25±0.05)、阴性对照组为(0.14±0.04)、治疗组为(0.17±0.03).治疗组较阴性对照组小鼠差异无统计学意义(P>0.05),而较阳性对照组过敏小鼠明显降低,差异有统计学意义(P<0.05).HE染色可见治疗组小肠绒毛中上皮细胞局灶性坏死、脱落,固有层炎症细胞浸润现象明显改善;IL-10表达增高.各组TGF-β表达无差异.结论 AdCTLA4Ig/Adα4β7修饰的耐受性DC对卵清蛋白过敏小鼠具有治疗作用,可以减轻肠道炎症反应,促进免疫耐受形成. Objective To investigate the therapeutic effects of AdCTLA4 Ig/Adα4β7-dendritic cell (DC) in the ovalbulmin (OVA) allergic BALB/c mice. Method Twenty-four Female BALB/c mice aged 4-6 weeks fed on the ovalbulmin-free feed were randomly divided into 3 groups: ( 1 ) treatment group: the OVA sensitized mice were injected with tolerogenic AdCTLA4- Ig/Adα4β7-DC after being challenged with OVA. (2) negative control group: mice were sensitized and challenged with normal saline. (3) positive control group: mice were sensitized and challenged with OVA. The level of the OVA-specific IgE in serum was measured by ELISA. The jejunal samples were observed histologically after HE staining. The level of IL-10 and TGF-β in murine intestine was detected by immunohistochemical and immunofluorescent methods, respectively. Result Compared with the positive group, caudal vein injection with AdCTLA4Ig/Adα4β7-DC in treatment group reduced the level of the OVA-specific IgE in the serum remarkably, inhibited the inflammatory reactions of the intestine and increased the expression of the IL-10 in intestine significantly.There was no significant change in the expression of TGF-β in each group. Conclusion This study demonstrated that caudal vein injection with tolerogenic AdTLA4Ig/Adα4β7-DC may be of potential research value in the treatment of food allergy by inducing immune tolerance in vivo.
