丙戊酸对白血病作用的研究进展

摘要 丙戊酸作为一种组蛋白去乙酰化酶抑制剂(HDACI),通过阻滞周期、激发凋亡、阻遏融合基因、影响归巢、调节免疫等多种途径影响白血病发生、发展。临床试验多用丙戊酸联合维甲酸、DNA甲基转移酶抑制剂,显示一定的抗白血病作用。但丙戊酸对白血病的作用机制、临床应用的有效性、安全性仍值得深入探讨。 As a kind of histone deacetylase inhibitor (HDACI) , valproic acid (VPA) influences the development of leukemia by cell cycle arresting , apoptosis induction, fusion gene repression, homing interfer- ence and immune- regulation. The clinical combination of all-trans retinoic acid or DNA methyltransferase in- hibitor with VPA has been demonstrated anti-leukemia effects. However, it needs further investigation on the anti-leukemia mechanism of VPA as well as its efficacy and safety in clinical application for leukemia patients.
