摘要 目的探讨丙戊酸(VPA)对耐三尖杉酯碱的HL-60细胞(HL-60/HT)耐药性的影响。方法选择递增压力培养法建立HL-60/HT细胞株。分为三组,Ara-C组、VPA组、VPA+Ara-C组,采用MTT法测定使用各组药物后敏感细胞、耐药细胞增殖情况的变化,同时测定丙戊酸对HL-60/HT细胞耐药性的改变,用流式细胞仪检测敏感的HL-60细胞及HL-60/HT细胞的细胞周期。结果丙戊酸与Ara-C合用可明显抑制HL-60、HL-60/HT细胞生长,抑制率高于丙戊酸或Ara-C单独的作用(q=1.37、1.51)。结论丙戊酸能够抑制HL-60、HL-60/HT细胞生长,可与Ara-C发挥协同作用,降低白血病细胞的耐药性。 Objective To explore the impact of Valproic Acid (VPA) on drug resistance of HL-60 cells(HL-60/HT) resistant harringtonine.Methods HL-60/HT Cell strain was established by incremental pressure culture and divided into 3 groups:Ara-C group,VPA group and VPA+Ara-C group.MTT method was used to determine the change of proliferation of sensitive cells and drug resistant cells after using drug.The change of VPA resistance in HL-60/HT cells was determinated simultaneously.The cell cycle of seneitive HL-60 cells and HL-60/HT cells were detected by flow cytometry. Results VPA in combination with Ara-C can obviously inhibit the growth of HL-60 and HL-60/HT cell,and the inhibition rate was higher than that of VPA or Ara-C alone (q=1.37,1.51).Conclusion VPA can inhibit the growth of HL-60 and HL-60/HT cell.It can play a synergistic role with Ara-C and reduce the drug resistance of leukemia cells.
