摘要
目的:研究制备抗皂角毒和表皮生长因子受体双功能抗体,用以导向皂角毒杀伤肿瘤细胞。方法:在制备EQ75、CY12.14单克隆抗体细胞株的基础上,通过杂交杂交瘤技术制备双功能抗体细胞株。结果:双功能抗体(EQ75×CY12.14)与单用皂角毒杀伤肿瘤细胞(A431)的IC50分别为10-9.6、10-8.4mol/L。结论:双功能抗体(EQ75×CY12.14)杀伤肿瘤细胞(A431),以IC50值计算,有效率为单用皂角毒的15倍左右。
Objective:To analyses
and prepare the bispecific monoclonal antibody of anti Saporin and anti epidermal growth
factor receptor(EGFR) which can enhance the effectiveness of tumor cell killing
activity.Methods:On the basis of establishing the cell lins which produce monoclonal antibody
against EGFR(EQ 75 ) and Saporin (CY12.14) hybri hybridoma technique was used to
construct the cell line of bispecific monoclonal antibody against Saporin and EGFR.Results:IC
50 of the bispecific monoclonal antibody and only saporin are 10 -9.6 mol/L and 10 -8.4
mol/L respectively in tumor killing cell (A 431 ).Conclusion:The effectiveness measured by IC
50 which bispecific antibody(EQ 75 ×CY12.14)tumor killing cell(A 431 ),is about 15 times
more than that of which only saporin dose.
