摘要
为了解大鼠和小鼠事丸EGFR的表达和分布规律及其与精子发生的关系,本试验应用BALB/c小鼠抗人A431细胞的EGFR单克隆抗体及ABC免疫组织化学方法,研究了大鼠和小鼠EGFR的表达和分布情况。免疫组织化学观察的结果显示:1.出生后,大鼠和小鼠睾丸中的间质细胞即表达EGFR。此时间质细胞体积小,细胞数量也少,所以EGFR的表达是有限。2.性成熟期,表达EGFR的睾丸间质细胞增多;此外,少数精原细胞、精母细胞及管周肌样细胞也表达EGFR。进入性成熟期后,睾丸内EGFR表达量增加。3.成熟期睾丸中,部分精原细胞和精母细胞分别表达EGFR或EGF,可通过自分泌或旁分泌机制,实现其局部调节。此机制有利于精子的发生。这些结果提示:在大鼠和小鼠睾丸中,EGFR的这一与性成熟有关的表达和分布规律与EGF是相似的。EGF通过EGFR作用于生精过程,与性成熟和精子发生关系密切。
In order to understand the pattern of epidermal growth factor receptor (EGFR) expression and distribution in the testis from rats and mice and to probe the relationship between this pattern and spermatogenesis, we used a BAL B/c mouse monoclonal antibody against the EGFR from the human A431 cell and the method of avidin-biotin-peroxidase complex (ABC) immunohistochemistry to investigate the state of EGFR expression and distribution in testis from rats and mice. The results showed that:1. After birth, the Leydig cells, limited in number, have undertaken EGFR expression, although maintained at low level in quantity. 2. During maturity, testes got a raise in the quantity of EGFR expressed by Leydig cells accompanied with the increase in the number. On the other hand, the additional EGFR expression appeared in a few of spermatogonia、spermatocytes and peritubular cells. 3. There is a local regulatory mechanism (autocrine and paracrine) in favor of spermatogenesis of mature testis by means of EGF and EGFR expression in Leydig cells、a few of spermatogonia and spermatocytes. These results suggested that the pattern of the EGF expression and distribution in rat and mouse is similar to that of EGF, and related to maturity too. EGF can exert its regulatory effect on spermatogenesis through EGFR,and has the close relation to maturity and spermatogenesis.
