摘要
结合1例色素浓重的脉络膜恶性黑色素瘤合并黑色素细胞瘤样本,对优化葡萄膜色素性肿瘤样本脱色素及免疫组化染色处理的方法进行探索。临床获得的脉络膜恶性黑色素瘤合并黑色素细胞瘤石蜡组织标本,分别使用不同浓度的高锰酸钾/草酸溶液进行脱色素处理,并进行Ki67免疫组化检测。结果表明:标本切片脱蜡后使用多聚甲醛或福尔马林后固定,再以质量分数为1%高锰酸钾/质量分数为2%草酸溶液进行脱色素处理能得到满意的HE(苏木素-伊红)染色结果;在脱色素之前进行抗原修复,继而经后固定后进行脱色素处理的组织切片,脱色素结果较好且保持了组织的抗原性。对于色素沉重牢固的葡萄膜色素性肿瘤,在脱色素处理前或者抗原修复后,增加后固定步骤,并使用较高浓度的高锰酸钾/草酸溶液,能得到较好的脱色素效果,且可保持组织的抗原性和切片的完整。
Aim:To optimize the method of bleaching and immunostaining on heavily pigmented choroidal tumor specimens,using the samples from an extremely heavy-pigmented choroidal melanoma arising from melanocytoma case.Method:Formalin fixed and paraffin embedded tissue were processed with or without postfixation and bleached by potassium permanganate/oxalic acid at different concentrations,followed by immunohistochemistry for Ki-67.Result:Those sections that were postfixed by paraformldehyde or formalin,then bleached with 1% potassium permanganate/2%oxalic acid showed satisfying result on HE staining.Antigen retrieval and postfixation before bleaching helped to achieve complete bleaching and maintain the antigenicity of tissue and integrity of sections.Conclusion:For extremely heavy-pigmented choroidal tumor,postfixation before bleaching or after antigen retrieval,and processing with higher concentration of potassium permanganate/oxalic acid,result in complete bleaching,good tissue antigenicity and section integrity.
