摘要
利用响应面法优化隐甲藻产DHA的培养基,在8种因素的单因素试验基础上,采用Plackett-Burman设计筛选出葡萄糖和胰蛋白粉为显著影响因子,维持其他组分浓度在低水平不变,对以上2因子爬坡逼近最大响应区域后利用中心组合设计和响应面分析得到最高点和主要因子的浓度。优化后的培养基组成为:葡萄糖30.54g/L,胰蛋白粉5.03g/L,酵母粉4g/L,甘油20g/L,不添加MgCl2和维生素溶液,其他组分同460培养基,在此培养基条件下,DHA产量达(907.54±1.02)mg/L,是优化前产量的2.48倍。
Medium composition of Crypthecodinium cohnii was optimized with response surface method. On the basis of single-factor experiments of eight factors, ingredients glucose and tryptone were determined as significant factors by using Plackett-Burman design. When the levels of other components were kept at the low concentration, the steepest ascent experiment was applied for factors glucose and tryptone to reach the largest region, highest point and the concentrations of those two significant factors were obtained by central composite experiments and response surface analysis. The optimized medium were as follows: glucose 30.54g/L, tryptone 5.03g/L, yeast 4g/L, glycerin 20g/L, without addition of MgCl2 and Vitamin solution, and the other components are the same as medium 460. With the optimized medium, DHA productivity was (907.54±1.02) mg/L, which was 2.48 times to control.
