摘要
目的:建立芪苓益肝颗粒中黄芪甲苷的含量测定方法。方法:采用高效液相色谱-蒸发光散射检测(HPLC-ELSD)法定量分析芪苓益肝颗粒中黄芪甲苷的含量,色谱柱Aglient Eclipse XDB C18(4.6 mm×250 mm,5μm),以乙腈-水(35∶65)为流动相,流速1.0 mL·min-1,柱温25℃;蒸发光散射(ELSD)检测器,漂移管的温度110℃,载气流速1.6 L·min-1。进样量10μL。结果:黄芪甲苷在0.498~9.96μg呈良好的线性关系,r=0.9998;平均回收率为98.05%,RSD 0.89%,在常温下8 h内稳定性较好。结论:HPLC-ELSD法简便快速,准确,重复性好,结果可靠,可作为芪苓益肝颗粒的质量控制方法。
Objective:To establish a determination method of the content of astragaloside in Qiling Yigan Granular formulation by HPLC-ELSD.Method:The content of astragaloside in Qiling Yigan Granular formulation was determined by HPLC-ELSD and Agilent ZORBAX SB-C18(4.6 mm × 250 mm,5 μm) column was used.The mobile phase was a mixture of acetonitrile-water(35∶ 65),the flow rate was 1.0 mL·min^-1,column temperature was of 25 ℃.The temperature of drift tube for ELSD was 110 ℃ and the flow rate of air was 1.6 L·min^-1.Result:The calibration curve of astragaloside was linear over the range of 0.498-9.96 μg(r = 0.999 8),the average recovery rate was 98.05%,RSD 0.89%.Conclusion:The method is convenient,sensitvive and accurate.It can be a method for quality control in production of Qiling Yigan Granular formulation.
