摘要
扩增片段长度多态性(Amplified Fragment Length Polymorphism,AFLP)技术具有多态性丰富,所需DNA量少,灵敏度高等优点,但流程繁琐,花费时间长。本文根据AFLP技术的特点,在酶切-连接和聚丙烯酰胺凝胶的制备,这两方面做了很大改进。采用优化的AFLP技术分析三倍体银鲫的DNA指纹图谱,不仅缩短了实验所需时间,而且获得了稳定清晰的扩增带型。
AFLP(Amplified Fragment Length Polymorphism) is characterized not only by the merits of highly polymorphoism,less DNA usage and higher sensitivity,but by the drawbacks of tedious procedure and time-consuming.In this paper,nearly each step of AFLP was optimized,especially in gel-making.DNA fingerprinting of triploid silver crucian carp(Carassius auratus gibelio Bloch) showed that using optimized AFLP can not only save much time but obtain reproductive results.
