摘要
用20个AFLP引物组合分析了黄颡鱼(Pelteobagru fulvidraco)雌、雄个体的遗传差异,共获得397个位点,雌性个体获得371个位点,多态位点比例为41.24%,雄性个体检测到391个位点,多态位点比例较高,达到93.35%,雌性和雄性的Nei's基因多样度分别为0.1418、0.3691,Shannon's信息指数为0.2094、0.5350,雌雄个体间的相似系数为0.8225~0.9889,遗传距离为0.0112~0.1775,用PHYLIP V3.6软件绘制了个体间的聚类图。另外,在4个AFLP引物组合中,共发现了6个位点在雌雄个体间表现出明显的扩增差异,其中4个位点显性条带个体为(?)性的比例为66.7%~80%,2个位点显性条带个体为(?)性的比例为100%。
Yellow catfish (Pelteobagrus fulvidraco) is widely distributed in China, especially in river and lake. Recently, it has become an important aquaculture species. It is generally matured at two years old and male is bigger than female. This study applied amplified fragment length polymorphism (AFLP) technique with 20 primer combinations to analyze the genetic structure of both male and female populations. 397 loci were obtained and the percentage of polymorphic loci was 41.24% in female population and 93.35% in male population. Nei' s gene diversity was 0. 1418 and 0. 3691 respectively. Shannon' s information index was 0.2094 and 0. 5350. Genetic similarity index ranged from 0. 8225 to 0.9889 and genetic distance was 0. 0112 to 0. 1775 between female and male. A dendrogram was constructed using PHYLIP V 3.6. 6 loci are identified from 4 primer combinations. Female genotype is dominant in 4 loci, which ratio is 66.7% - 80% ; and male ratio is 100% in dominant individuals of 2 loci.
