摘要
目的探讨P-糖蛋白(P-gp)、谷胱甘肽转移酶(GST-π)、DNA拓扑异构酶-Ⅱ(Topo-Ⅱ)、人类表皮生长因子受体-2(HER-2)在乳腺癌组织中的表达及其与含多西他赛方案化疗疗效的关系。方法采用SP免疫组化法检测54例患者肿瘤组织标本P-gp、GST-π、Topo-Ⅱ、HER-2的表达,给予多西他赛联合蒽环类或铂类化疗方案进行化疗,分析蛋白表达与化疗疗效的关系。结果 P-gp、GST-π、Topo-Ⅱ、HER-2阳性表达率分别为55.6%、18.5%、85.2%、81.5%,P-gp和HER-2、GST-π和HER-2的表达具有明显相关性(P<0.05);化疗总有效率为44.4%,P-gp、GST-π、Topo-Ⅱ、HER-2表达水平与化疗疗效无明显相关性(P>0.05)。结论 P-gp、GST-π、Topo-Ⅱ、HER-2的表达与含多西他赛方案化疗疗效间无明确相关性,其与化疗疗效的关系及是否可以作为筛选化疗药物的指标,有待于进一步研究。
Objective To explore the correlation between the protein expression of P-gp、GST-π、Topo-Ⅱ、HER-2 and the therapeutic effectiveness in breast cancer patients treated with docetaxel-based combination chemotherapy.Methods Detected P-gp、GST-π、Topo-Ⅱ、HER-2 protein expression in the specimens of 54 breast cancer patients by S-P immunohistochemistry,and the patients were treated with docetaxel combined with anthracycline or platinum chemotherapy.Analyzed the relationship between the protein expression and chemotherapy efficacy.Results The positive expression rate of P-gp、GST-π、Topo-Ⅱ、HER-2 was 55.6%、18.5%、85.2%、81.5%,respectively,and there was significant correlation between the expression of P-gp and HER-2,GST-π and HER-2（P〈0.05）.The total effective rate was 44.4%.The positive expression level of P-gp、GST-π、Topo-Ⅱ、HER-2 in breast cancer was not correlated definitely with docetaxel-based combination chemotherapy（P〉0.05）.Conclusion There is no interrelated relationship between the expression of P-gp、GST-π、Topo-Ⅱ、HER-2 and the chemotherapy effectiveness of docetaxel combined with anthracycline or platinum,and further investigation will be need to do in order to make clear there correlations with chemotherapy efficacy and to confirm whether they can serve as a valuable marker in selecting chemical drugs.
