摘要
目的研究DNA拓扑异构酶-Ⅱα(Topoisomerase-Ⅱα,Topo-Ⅱα)基因在人脑胶质瘤中的表达改变及其对胶质瘤U87MG细胞凋亡的影响。方法 Real-time PCR检测25例脑胶质瘤及相应癌旁组织中Topo-ⅡαmRNA的表达。将Topo-Ⅱα基因特异性的siRNA转染U87MG细胞。转染后,Western Blot检测转染后Topo-Ⅱα蛋白的表达,双标流式细胞法检测细胞凋亡率。结果与癌旁组织相比,Topo-ⅡαmRNA在脑胶质瘤组织中表达明显上调。转染后,U87MG细胞中的Topo-Ⅱα蛋白表达明显降低,同时,凋亡率明显增加。结论 Topo-Ⅱα基因在脑胶质瘤中存在表达异常,其表达降低能够促进细胞凋亡。
[ Objective ] To research the relationship of the expression changes of Topo-IIα gene in brain glioma, and the influence to apoptosis in U87MG cells. [ Methods ] 25 cases of brain glioma and peri-cancerous normal tissues were selected to detect the expression of Topo-IIα mRNA by Real-time PCR. Specific siRNA for Topo-IIα gene was transfected to U87MG cells. After transfection, the expression of Topo-IIα protein was detected by western blot, and the apoptosis of the transfected cells was detected by flow cytometry. [ Results ] The expression of Topo-IIα mRNA presented an obvious increase in brain glioma, compared with its expression in peri-cancerous normal tissues. After transfection, the expression of Topo-IIα protein in the transfected U87MG cells was down-regulated significantly and the apoptosis increased significantly. [ Conclusions ] The expression of Topo-IIα gene in brain glioma changes abnormally. The low expression of Topo-IIα gene can increase the apoptosis of U87MG ceils.
