摘要
目的研究Survivin、EGFR和VEGF在肝细胞癌中的表达情况,为肝癌预后判断和靶向治疗提供理论依据。方法应用免疫组织化学PV6000法检测81例人肝细胞癌组织、癌旁组织Survivin、EGFR和VEGF的表达并分析其与相关临床病理特征的关系。结果肝细胞癌组织与癌旁组织Survivin的阳性率无显著差别;肝细胞癌组织EGFR、VEGF的阳性率分别为49.4%、63.0%,显著高于癌旁组织的阳性率21.0%、44.4%(P<0.05)。Survivin的表达与肿瘤的TNM分期和肿瘤直径有关(P<0.05);EGFR的表达与病理分化和术后复发转移有关(P<0.05);VEGF的表达与TNM分期、术前局部转移及术后复发转移有关(P<0.05)。EGFR与VEGF表达呈正相关(r=0.307,P=0.007)。TNM分期、Survivin、EGFR是肝细胞癌的独立预后因子(P<0.05)。结论肝细胞癌组织Survivin、EGFR和VEGF表达水平较高,其中Survivin、EGFR可以作为肝细胞癌的预后因子。EGFR与VEGF表达正相关,对研发多靶点药物或联合使用多种靶向药物有一定指导意义。
Objective To investigate the expression of survivin,EGFR and VEGF in hepatocellular carcinoma(HCC),so as to provide theoretical basis for prognosis and molecular targeted therapy.Methods The expression of the proteins mentioned above in 81 HCC tissues and their corresponding non-cancerous adjacent liver tissues were detected by PV6000 immunohistochemistry method,and the follow-up informations of the patients were investigated.Results There was no significance between the expression rate of survivin in HCC tissues and non-cancerous adjacent liver tissues.The expression rate of EGFR and VEGF in HCC tissues was 49.4% and 63.0%,respectively,which was significantly higher than that in non-cancerous adjacent liver tissues(21.0% and 44.4%,respectively)(P〈0.05).The expression of survivin was related to the TNM stage and the size of tumors(P〈0.05).The expression of EGFR was related to the histological grade and the metastasis after resection(P〈0.05).The expression of VEGF was related to the TNM stage,the local metastasis before resection and the metastasis after resection(P〈0.05).There was a significant correlation between the expression of EGFR and VEGF in HCC tissues(r=0.307,P=0.007).The TNM stage,the expression of survivin and EGFR were independent risk factors affecting the prognosis of patients with HCC(P〈0.05).Conclusion The expression rates of survivin,EGFR and VEGF were high in HCC tissues,of which survivin and EGFR could be used as the predictors of the prognosis for the patients with HCC.The correlation between the expression of EGFR and VEGF was useful for developing multi-target anti-tumor drugs or combining using molecular targeted drugs targeting EGFR and VEGF.
