摘要
目的分析接种A(H1N1)疫苗人群血清的中和抗体对2009年A(H1N1)的抗病毒作用和相关病毒的交叉免疫保护作用。方法利用MDCK细胞的细胞病变检测接种A(H1N1)疫苗的人血清和未免疫的对照血清对甲型流感病毒A/Brisbane/10/2007(H3N2),A/Brisbane/59/2007(H1N1),A/California/07/2009(H1N1)和A/Shenzhen/406H/2006(H5N1)等病毒的中和作用。结果通过细胞病变观察,证实接种A(H1N1)疫苗的人血清稀释度为1∶40时,30份免疫血清可以中和H3N2(Brisbane),H1N1(Brisbane)和H1N1(CA7)而不产生细胞病变,中和保护率分别均为100%,而相同稀释度的未免疫对照血清的中和保护率分别为100%,100%,40%;而当稀释度为1∶400时,30份免疫血清分别有13,20和21例未出现细胞病变,中和保护率分别为43%,67%,70%,10份对照血清的中和保护率分别为80%,70%,0%。两种稀释度的免疫血清和未免疫对照血清均不能中和H5N1引起细胞病变,中和保护率均为0%。结论接种2009年A(H1N1)疫苗可以诱导能中和CA7 H1N1的抗体产生,但该中和抗体对H3N2(Brisbane),H1N1(Brisbane),H5N1(SZ)高致病禽流感病毒等甲型流感病毒无交叉保护作用。
Objective To investigate whether year 2009 A(H1N1) vaccine induces anti-virus activity against A(H1N1) and cross-reactive immune response against related A type influenza viruses at cell level.Method A/ Brisbane /10 /2007(H3N2),A / Brisbane /59 /2007(H1N1),A /California /07 /2009(H1N1) or A /Shenzhen /406H /2006(H5N1) were preincubated with 30 sera vaccinated with year 2009 A(H1N1) or 10 unvaccinated sera from donors respectively,and then used to infect MDCK cell,neutralizing activities were defined according to Cytopathic Effect(CPE).Results When serum dilution was 1∶40,all of 30 sera were CPE negative when incubating with H3N2(Brisbane),H1N1(Brisbane) and H1N1(CA7) respectively,and the percentage of positive neutralization reaction wereall 100%,for the unvaccinated sera,the percentage were 100%,100%,40% accordingly;while there were 13,20,21 vaccinated sera showing CPE negative results at dilution of 1:400,with percentage of positive neutralization reaction 43%,67%,70% accordingly.As for the 10 control sera,the percentage were 80%,70%,0%.In addition,as for H5N1(SZ),the results of all 30 vaccinated and 10 control sera were all CPE negative at both serum dilution of 1∶40 and 1∶400,so the percentage were 0%.Conclusions The neutralizing antibodies raised in all 30 sera vaccinated with year 2009 A(H1N1) can neutralize CA7 H1N1,but cannot cross-react to H3N2(Brisbane),H1N1(Brisbane) and H5N1(SZ).
