摘要
目的 探讨旋毛虫与血吸虫之间交叉免疫的分子机制 ,并寻找有效的日本血吸虫疫苗候选分子。方法 以旋毛虫感染鼠血清为探针 ,筛选日本血吸虫成虫 c DNA文库 ,对获得的阳性克隆插入片段进行 PCR扩增。结果 共筛选出 11个阳性克隆 ,其插入的 c DNA片段大小在 1.4~ 5 .0 kb之间 ,其中 1.4kb片段 2个 ,5 .0 kb片段 9个。结论 获得的阳性克隆插入基因片段可能为编码旋毛虫与日本血吸虫共同的抗原基因 。
ObjectivesTo explore the molecular mechanism of cross reactivity between Trichinella spiralis (Ts)andSchistosoma japonicum (Sj) , and to find out effective vaccine candidate antigens against schistosomiasis japonica. Methods Sj adult worm cDNA library was screened using sera of mice infected with Tsand the inserts of positive clones were specifically amplified by PCR. Results Eleven positive clones were obtained after three rounds of immunoscreening. The size of these inserts ranged from 1 4 to 5 0 kb. Two of these fragments were 1 4 kb, and nine were 5 0 kb. Conclusions The positive clones may be the genes encoding the same antigens ofTs and Sj, and their expressed products could be used as vaccine candidates forSj infection.
