摘要
以黑穗醋栗花为试验材料,利用紫外分光光度计和凝胶电泳法比较了Trizol法、硼砂-SDS法、CTAB法、植物RNA提取试剂盒等4种方法提取总RNA的质量和纯度。结果表明,利用硼砂-SDS法提取的RNA,28S和18S两条带型清楚,两条带之间无弥散现象,且纯度最高,D260/D280达到2.06,经计算得出总RNA产率达61.60μg·g-1,反转录条件下可得500~1000bp之间的弥散cDNA条带,进一步证明硼砂-SDS法提取得到的总RNA具有很高的纯度,可以满足进一步分子生物学研究的需要。
CTAB method,Borax-SDS method,Trizol method,Plant RNA out Kit method were employed to extract total RNA in flower of the ripe blackcurrant.Through UV spectrometer and gel electrophoresis indicated such as the total RNA extracted from four kinds of quality and purity.The results showed thatthe use ofBorax-SDS extraction ofRNA,28S and 18S two banding patterns clearly between the two with no dispersion phenomenon,and the highest purity,D260/D280 to 2.06,calculated that the total RNA yield of61.60 μg·g-1,reversed the transcriptase underthe conditions of the available 500-1 000 bp dispersion among the cDNA bands,the further evidence of Borax-SDS method of total RNA were extracted with high purity,to meetthe need forfurthermolecularbiology research.
