摘要
目的:建立一种可用于莽草酸及其衍生物定性检测的高灵敏度荧光薄层色谱方法。方法:含有莽草酸及其衍生物的甲醇溶液经硅胶薄层层析展开后喷洒0.1%的高锰酸钾-1%硫酸水溶液,105℃加热20min后在365nm下观测荧光。结果:莽草酸及其单、双、三取代后的衍生物在365nm下均可观测到蓝绿色荧光,检出限可达0.7mg(4.02nmol)。结论:本研究为莽草酸及其衍生物的定性鉴定提供了一种快速、灵敏、简便的方法,可广泛用于莽草酸及其衍生物的分析和检测。
Objective:To establish a highly sensitive fluorescent thin-layer chromatography method for detection of shikimic acid and its derivatives. Methods:After development,the silica gel containing shikimic acid and its derivatives were sprayed by 0.1% solution of KMnO4 in 1% H2SO4 aqueous solution,and then heated in 105 ℃ for 20 min,then observed under 365 nm. Results:Under 365 nm,blue-green fluorescence could be observed for shikimic acid and all of its derivatives,either mono-,di-,or tri-substituted derivatives. The lowest detective limit was as low as 0.7 mg(4.02 nmol). Conclusion:This study provides a rapid,sensitive and simple method for the detection of shikimic acid and its derivatives. It can be used for the analysis and testing of shikimic acid and its derivatives.
