摘要
为了优化八角茴香中莽草酸渗漉法提取工艺参数,建立提取物TLC法.通过单因素试验和L9(34)正交试验设计-综合评分法,以莽草酸和干膏率为评价指标,采用渗漉法、UV法考察乙醇浓度、料液比、提取时间、药材粒度、渗漉速度与渗漉次数对工艺条件的影响,并对莽草酸进行TLC鉴别.结果发现取八角茴香粉末(80目),加料液比1:15 g/mL的60%乙醇渗漉提取12 h,流速1 mL/min,渗漉1次,测得3批样品中莽草酸的平均含量为9.40%;该样品的TLC鉴别斑点清晰,分离度较好,与文献一致.因此认为本实验优化的工艺条件操作简捷,稳定可行,具有实际应用价值.
To optimize the extraction parameters of shikimic acid via percolation method while establishing TLC method of the extraction from Star Anise.The single factor experiment,L9(34)orthogonal test design and comprehensive score method as well as the method of diacolation and UV were used to estimate the influence of diacolation process conditions under ethanol concentration,solid-liquid ratio,extraction time,medicinal material granularity,diacolation speed and times while regarding the rate of shikimic acid and dried extract as the evaluation index.Meanwhile,shikimic acid of the extracts was identified by TLC.Results show that Anise powder(80 mesh)was percolated for 12 hours with solid-toliquid ratio 1:15(g/mL)with 60%ethanol at a flow rate of 1 mL/min for one time.The average content of shikimic acid in 3 batches of samples was 9.40%.The clear TLC identification of the sample and good separated degree were in accordance with the literature.Therefore,it is considered that the optimized technological conditions in this experiment are simple,stable and practical.
