摘要
以高效液相色谱法分别测定了6种碳源和8种氮源对莽草酸发酵菌生产莽草酸能力的影响,结果表明:碳源中葡萄糖和果糖对莽草酸产量影响最大,产量分别达到1.43 mg/L和1.50 mg/L;氮源中蛋白胨对莽草酸的贡献最大,达134.58 mg/L.最终确定后续培养基的成分分别为:葡萄糖、蛋白胨、磷酸氢二钠、磷酸氢二钾、氯化钠.
The production of shikimic acid was assayed with high performance liquid chromatography while six kinds of carbon sources and eight sorts of nitrogen sources were added into the minimal media respectively.The result showed that the production of shikimic acid was 1.43 mg/L fermented in the medium added glucose,and the production was 1.50 mg/L in the medium dissolved fructose.This two carbon sources contributed to more shikimic acid than others.Glucose was,however,determined as the carbon source of the designed medium when the price factor was considered.In nitrogen sources,peptone contributed to the highest shikimic acid production.The production reached 134.58 mg/L.And thus peptone was the optimal nitrogen source.Therefore,the ingredients of the designed medium were glucose,peptone,disodium hydrogen phosphate,potassium dihydrogen phosphate,and sodium chloride.
