摘要
目的:确定响铃草抗炎镇痛活性部位,建立该部位质量控制的方法。方法:小鼠耳廓肿胀法筛选抗炎活性部位,醋酸扭体法及热板法筛选镇痛活性部位。并以大豆皂苷元B为指标性成分,5%香草醛-高氯酸为显色系统,采用分光光度法在568nm波长处测定活性部位中总皂苷的含量。结果:响铃草正丁醇萃取部位具有明显的抗炎镇痛活性。大豆皂苷元质量在60.6~109.08μg范围内与吸光度线性关系良好,回归方程为Y=0.0103X-0.2908,r=0.9991。结论:响铃草正丁醇萃取部位为响铃草的抗炎镇痛部位;分光光度法测定该部位总皂苷的含量,操作简单,所得实验结果稳定、可靠,可作为响铃草抗炎镇痛活性部位质量控制的方法。
Objective:To confirm the effective fraction on anti-inflammatory and analgesia,and to establish the methods of quality control for the effective fraction of Crotalaria ferruginea Grah..Methods:For effective fraction,analgesic effects was employed hot-plate procedure and the mouse torsion body induced by acetum method;the anti-inflammatory effect were optimized by the mouse pinna swelling induced by xylene. The content of total triterpene saponins was measured by spectrophotometry with 5% vanillin and perchloric acid as color-developing agent,and 12-oleanene-3β,22β,24-triol as the standard sample.Results:n-Butanol-extracted fraction showed obvious effection on anti-inflammatory and analgesia.The calibration curve was in the range of 60.6-109.08 μg,and the curve was Y=0.0103X-0.2908,r=0.9991.Conclusion:n-Butanol-extracted fraction has been proved to be effective for anti-inflammatory and analgesia.The spectrophotometry mthod is suitable for the determination of total triterpene saponins of the n-butanol-extracted fraction,and good for quality control.
