高效液相色谱法测定大剂量甲氨蝶呤肿瘤化疗患者的血药浓度 被引量：4

摘要 目的:建立大剂量化疗时人血浆中甲氨蝶呤浓度的测定方法。方法:以Kromasil C18(4.6mm×250mm,5μm)为色谱柱,流动相为0.05%磷酸盐缓冲液(pH6.8)-甲醇(80-20);柱温45℃;流速0.8mL·min-1;检测波长306nm。结果:在0.08～100mg·L-1范围内,线性关系良好,r=0.9999,最低检测为0.02mg·L-1,平均加样回收率99.2%,日内及日间RSD均小于5%。结论:该方法简单快捷,灵敏度高,结果准确,适用于大剂量化疗时甲氨蝶呤血药浓度监测。
