摘要
目的:探讨益智仁乙醇提取物(Alpiniae oxyphyllae ethanolic extract,AOE)对6-羟基多巴胺(6-hydroxydopamine,6-OH-DA)在PC12细胞上引起的神经损伤的保护作用及其机制。方法:采用4-甲基偶氮唑蓝比色法(MTT法)和乳酸脱氢酶释放法(LDH法)检测PC12细胞的活力;DAF-FMdiacetate荧光染色检测细胞内一氧化氮(NO)的含量;采用免疫印迹法检测诱导型一氧化氮合酶(iNOS)的蛋白表达水平。结果:AOE能够剂量依赖性的保护PC12细胞的活力,减少6-OHDA引起的PC12细胞内LDH释放。作用机理研究证明AOE能够降低6-OHDA刺激的PC12细胞内NO含量增加。免疫印记杂交的结果显示AOE能够降低iNOS的表达。结论:AOE能保护6-OHDA诱导的PC12细胞损伤,其作用机制可能与降低NO的产生和iNOS的表达。该研究中证实的AOE神经保护效应对帕金森病的防治可能具有一定的作用。
Present study was planned to explore the neuroprotective effect of Alpiniae oxyphyllae ethanolic extract(AOE) and to elucidate its underlying mechanism of action.6-hydroxydopamine(6-OHDA),selective neurotoxin against dopaminergic neurons,was used to induce neuronal cell damage in PC12 cells.Pretreatments of AOE(3、6、12、25、50μg/ml) increased the viability of 6-OHDA-damaged PC12 cells and decreased LDH leakage in a dose dependent manner.Mechanistic study showed that anti-oxidative(inhibition of NO production and iNOS expression in PC12 cells in vitro) activities of AOE may be involved in its neuroprotective effect in 6-OHDA-induced DA neuron damage.The present study suggests that Alpiniae oxyphyllae may be useful in the treatment of Parkinson's disease.
