摘要
目的:为证明构成内毒素(LPS)的主要核心部分类脂A(LipidA)在LPS直接损伤内皮细胞中所起的作用。方法:分离培养人肾小球内皮细胞(HGVEC),把细胞分为正常对照组、LPS时间梯度组、LPS浓度梯度组和LipidA浓度梯度组。线粒体脱氢酶(Mitochondriadehydrogenase,MtDH)活性采用MTT法测定。结果:时间梯度组:1μg/mlLPS使内皮细胞线粒体脱氢酶活性呈进行性下降,在12h时比正常对照组下降显著(P<0.01)。浓度梯度组LPS在0.9μg/ml使线粒体脱氢酶活性显著下降,而LipidA在0.6μg/ml处即可出现显著下降。结论:LPS和LipidA均可直接损伤HGVEC,使其线粒体脱氢酶活性下降,LipidA的损伤作用强于LPS,证明LipidA在LPS引起的内皮细胞直接损伤中起主要作用。
Objective: To investigate the injurious effects of LPS and lipid A on vascular endothelial cells (VECs). Methods: Cultured human glomerular vascular endothelial cells (HGVECs) were divided into the control group and the groups treated with LPS in different time durations, the groups treated with LPS in different concentrations and the groups treated with lipid A in different concentrations. The activity of mitochondrial dehydrogenase was measured with MTT. Results: LPS 1 μg/ml inhibited significantly the activity of mitochondrial dehydrogenase ( P <0.01). The inhibiting concentration was lower in lipid A (0.6 μg/ml) than in LPS (0.9 μg/ml). Conclusion: Both LPS and lipid A possess direct injurious effects on HGVECs and inhibit the activity of mitochondrial dehydrogenase. The inhibition is more powerful in lipid A than in LPS. The finding suggests that lipid A plays a major role in the injurious effects on VECs.
