摘要
在芡实多酚的柱层析纯化过程中,为了选择合适的吸附剂,本研究分析了聚酰胺、D101树脂和中性氧化铝三种吸附剂对芡实多酚的吸附特性。结果显示,聚酰胺、D101树脂和中性氧化铝三种吸附剂对溶液中芡实多酚的平衡吸附量均较高,在多酚初始浓度为0.5mg/mL时,平衡吸附量依次为34.48、33.21、35.49mg/g。其中,D101树脂对溶液中芡实多酚的吸附具有较高的选择性,是分离纯化芡实多酚较理想的吸附剂。芡实多酚经D101树脂吸附后,用pH=5,50%丙酮水溶液(v/v)洗脱,即可将其中91.84%的芡实多酚洗脱下来。洗脱液经减压浓缩、干燥后,得纯化芡实多酚样品,其中总酚含量达81.38%。
In order to choose a proper adsorbent for purification of the polyphenol in gorgon nut (GNPP), neutral alumina,polyamide(PA) and DI0! resin were chosen, and the adsorption characteristics of three adsorbents to GNPP in solution were analyzed and investigated in this test.The results showed that neutral alumina, polyamide (PA) and DIO! resin all had highly equilibrium adsorption quality(Qe).When the initial concentration was 0.5mg/mL, the Qe was 35.49,34.48,33.21mg/g, respectively. Moreover, D101 resin had the highest selective adsorption to GNPP in solution,so was regarded as the perfect adsorbent for GNPP purification.In GNPP purifition,the 91.84% GNPP adsorbed by D101 resin can be desorbed with pH5, 50% acetone aqueous (v/v).The effluent was concentrated and dehydrated in vacuum and the purified GNPP sample was attained, in which the total polyphenols content was 81.38%.
