摘要
目的:通过改进离体心脏灌流试验,延长家兔离体心脏存活时间。方法:健康大耳长白家兔10只随机分为对照组与实验组,每组5只;通过改良Krebs-Henseleit液(K-H)配方与部分术式操作,比较两组的体外存活时间。结果:与对照组比较,改进组家兔离体心脏在体外稳定有节律跳动时间明显延长(P<0.01)。结论:改良部分手术操作以充分暴露离体心脏,加之去掉K-H配方之Na2EDTA而减少与Ca2+络合以相对提高有效Ca2+浓度,伴同时增加镁离子浓度,以共同改善心肌收缩功能和延长离体兔心脏存活时间。此简捷模型宜用于专业实验教学与筛选心脏药物等研究。
Objective: To prolong survival time of the isolated rabbit heart by improving perfusion experiment for isolated rabbit hearts.Methods:10 rabbit were divided into control group and treated group,5 rabbits/group.Observe the prolonging survival time after the modified Krebs-Henseleit and the some operation.Results: Compared to the control group,the prolonging survival time of treated group was over 8 hours in vitro(P<0.01).Conclusions:the main mechanism of prolonging survival time deals with the formulation modification and the improvement of some operation and the increase in Ca2+and Mg2+.The model was useful to the teaching and research in the effects of drugs on the rabbit hearts.
