摘要
[目的]筛选微生物絮凝剂产生菌的培养条件,并对其絮凝成分进行分析。[方法]从成都市土壤中筛选分离了1株具有稳定高效的微生物絮凝剂产生菌MB-7。考察了碳源、氮源、pH值、温度、培养时间和摇床转速对絮凝效果的影响,对培养条件进行优化。最后,对其活性成分分布及成分进行研究。[结果]该菌株产絮凝剂的最佳培养条件为:碳源为淀粉,氮源为硫酸铵,培养时间为72h,pH值为7.0,培养温度为30℃,摇床转速为160r/min。在最佳条件下,该菌株对4%高岭土悬浊液的絮凝率达到94.5%。该菌絮凝活性物质主要分布在发酵液中,主要成分为多糖,含量高达85.7%。[结论]该研究可为开发高效、无毒、无二次污染和能生物降解的水处理剂奠定基础。
[Objective] The research aimed to screen culture condition of bioflocculant-producing and analyze the component of microbial flocculant. [Method] A flocculant-producing strain MB-7 was isolated. The effects of different culture condition such as carbon source,nitrogen source,pH value,temperature,culture time and rotation speed on flocculation rate were studied. [Result] The optimum culture condition of flocculant produced by strain MB-7 was as follow:carbon source was starch,nitrogen source was ammonium sulfate,the optimum culture time was 72 h,pH value was 7.0,culture temperature was 30 ℃ and rotation speed was 160 r/min. The flocculating rate of strain MB-7 was 94.5% against 4% Kaolin clay suspension. The main active components of flocculant presented in the fermentation broth,and the main components of MBF-7 was polysaccharide,the content was 85.7%. [Conclusion] The study can lay basis for developing high efficiency,no poison,no second pollution and bio-degradable water treatment agent.
