摘要
[目的]优化果胶酶法提取叶下珠(Phyllanthus urinaria L.)中总黄酮的工艺。[方法]以蒸馏水为提取剂,在提取过程中加入果胶酶,以黄酮提取率为指标,采用正交试验考察提取温度、pH值、料液比、果胶酶浓度和提取时间对叶下珠黄酮得率的影响。[结果]采用果胶酶法提取叶下珠中黄酮的最佳工艺条件为:以pH值为5的蒸馏水为提取剂,料液比1∶30(W/V),果胶酶浓度为2.0g/L,在温度为55℃条件下提取150min。在此优化条件下,叶下珠中黄酮的提取率较高,为1.469%。[结论]该研究可为进一步开发利用叶下珠植物资源提供科学依据。
[Objective] The research aimed to optimize the extraction process of flavone in Phyllanthus urinaria L.based on pectinas method.[Method] The flavone in Phyllanthus urinaria L.was extracted by using wateras the extraction agent,and pectinas was added during extraction.The orthogonal test was used to study the effects of extraction temperature,pH value,the ratio of material to liquid,the concentration of pectinase and extraction time on the extraction ratio of flavone in Phyllanthus urinaria L..[Result] The results showed that the optimized extraction condition of flavone in Phyllanthus urinaria L.By pectinas method was as follow:the pH value of extraction agent was 5,the ratio of material to liquid was 1∶30,the concentration of pectinas was 2.0 g/L,the extraction temperature was 55 ℃ and the extraction time was 150 min.Under the optimized condition,the extraction ratio of flavone was high of 1.469%.[Conclusion] The study can provide the scientific basis for deeply exploring and utilizing the Phyllanthus urinaria L.Resources.
