摘要
[目的]建立从锁阳中富集儿茶素的方法,并对儿茶素含量进行测定。[方法]用50%乙醇对锁阳进行超声提取,提取物经大孔吸附树脂对儿茶素进行富集,并对儿茶素富集部位进行含量测定。HPLC工作条件为:色谱柱C18(250.0mm×4.6mm,5μm);流动相为乙腈∶0.2%乙酸15∶85(V/V);柱温为室温;流速1ml/min,检测波长280nm。[结果]锁阳中的儿茶素主要集中在35%乙醇洗脱部分;阴性对照品与样品分离良好,儿茶素的线性范围为0.005~0.040mg/ml(r=0.9999),回收率为98%,RSD为0.658%。[结论]用大孔吸附树脂可从锁阳提取物中高效地分离纯化富集儿茶素;HPLC检测方法可行,测定误差小,结果较准确,重现性好。
[Objective] To study the isolation and purification methods of catechin of Cynomorium songaricum Rupr.and establish a HPLC method for the determination of catechin.[Methods]The dried Cynomorium songaricum Rupr.powder was extracted with 50% EtOH and purified via column chromatography on macroporous adsorbent resin.Using the reversed-phase HPLC determine the content of catechin of above fraction.The qualification of HPLC:C18column(250.0 mm×4.6 mm,5 μm),with mobile phase consisted of acetonitrile:0.2%acetic acid(V/V)(15∶ 85).The column temperature was room temperature.The flow rate was 1 ml/min and the detection wavelength was 280 nm.[Results]Catechin is concentrated the 35% EtOH eluting fraction.The sample and the negative control were separated well by HPLC.The linear ranges of catechin was 0.005-0.040 mg/ml(r=0.999 9).The average recoveries of catechin was 98%(RSD=0.658%).[Conclusion] This method is simple and rapid to isolate and purify the catechin from Cynomorium songaricum Rupr.and,the feasible quantitative analysis method with small determinate error,accurate result and good reproducibility.
