摘要
[目的]研究转甘露醇-1-磷酸脱氢酶基因(mtlD)花生幼苗的抗盐性。[方法]通过根癌农杆菌介导将外源mtlD导入花生,通过检测植株的鲜重、高度等指标来判断转基因花生的抗盐性。[结果]PCR Northern杂交显示,mtlD已成功整合到染色体上;转基因植株的鲜重、高度、甘露醇-1-磷酸脱氢酶的活性都明显高于对照植株。[结论]mtlD基因在花生中的表达增强了转基因花生的抗盐性。
[Objective] The salt tolerance of transgenic peanut was studied.[Method] The mtlD gene was transformed into peanut with agrobacterium.The salt resistance of transgenic peanut was determined by detecting the plant fresh weight,height,conductivity and mannitol-1-phosphate dehydrogenase activity.[Result] PCR and Northern blot analysis indicated that mtlD gene had been integrated into genome of the peanut and expressed in transgenic peanut plants.The values of fresh weight,height and mannitol-1-phosphate dehydrogenase activity increased significantly compared with the control.[Conclusion] The expression of the mtlD gene could enhance the salt tolerance of the peanut.
