摘要
目的探讨氧自由基对培养犬呼吸道上皮细胞合成释放小分子活性物质一氧化氮(NO),内皮素1(ET1),前列腺素E2(PGE2])和血栓烷素(TXA2)的影响。方法培养的犬呼吸道上皮细胞与黄嘌呤(26.3μmol·L1)及黄嘌呤氧化酶(17u·L1)孵育4h后,应用酶联免疫、放射免疫及Gries还原法,测定条件培养液中NO,ET1,PGE2和TXA2含量。结果氧自由基使犬呼吸道上皮细胞ET1和TXA2释放量分别增加12倍和10倍;NO释放量减少60%;同时使PGE2的基础分泌量,组织胺刺激分泌量和花生四烯酸刺激分泌量均减少约73%~85%。结论氧自由基引起支气管上皮细胞参与调节支气管平滑肌舒缩的小分子活性物质分泌紊乱。
Objective To investigate the effects of oxygen free radicals on release of enogenous active molecules of cultured canine airway epithelial cells(CAEC),such as nitric oxide (NO),endotheline 1(ET 1),prostaglandin E 2(PGE 2) and theromboxane(TXA 2).Method Cultured CAEC were incubated with xanthine(26.3 μmol·L 1 )plus xanthine oxidase (17u·L 1 )(X XO)for 4 hours.The contents of NO,ET 1,PGE 2 and TXA 2 were measured by means of Griess assay,immunoradiation assay and ELISA,respectively.Results Treatment of X XO increased the release of ET 1 and TXA 2 by 12 and 10 folds, respectively. However,the release of NO and PGE 2 from CAEC were significanty reduced 60% and 73%~85% by X XO. Superoxiduse dismutase (100 u·L 1 )and catalase (200 u·L 1 )could block the effects of X+XO.Conclusion Oxygen free radicals leads disordered release of NO,ET 1,PGE 2 and TXA 2 airway epithelial cells,which is involved in the constringency of bronchus smooth muscle.
