摘要
目的 :探讨低压缺氧时与内皮细胞相关的一氧化氮 (NO) ,内皮素 (ET- 1)、肾上腺髓质素 (ADM)的变化 ,了解低压缺氧的损伤机制。方法 :将 30只雄性 Wistar大鼠 ,随机分为 5组 :对照组、低压缺氧后 30 min,6 h组、使用 N-甲基 - L-精氨酸 (L- NAME)保护后 30 min和 6 h组 ,每组 6只。实验组大鼠在低压舱内模拟上升至 80 0 0 m、停留 30min,重复 1次。分别于缺氧后 30 min和 6 h处死大鼠取血及心脏、肺组织 ,测定 NO,ET- 1,ADM及半定量测定心脏及肺组织 e NOSm RNA,i NOSm RNA。结果 :低压缺氧后大鼠血中 NO,ET- 1及心脏和肺组织 i NOSm RNA显著增高 ,ADM,e NOSm RNA显著降低。注射 L- NAME后可明显减少 NO,ET- 1,ADM的变化值。结论 :NO在低压缺氧后对 ADM,ET-
AIM:To study changes of NO,ET-1,ADM and mRNA expression of eNOS,iNOS after acute hypoxia.MEHHODS:Thirty conscious Wistar rats were randomly divided into 5 groups:1 control group and 4 experimental groups. Two of experimental groups were injected N-Methyl-L-Arginine(L-NAME,100 mg/kg) before experiment. The experimental groups underwent 8000 m altitude exposure in hypobaric chamber for 30 min,and repeat it after 0 m altitude. The rat blood and heart and lung were taken 30 min,6 h after exposure and NO,ET-1,ADM and eNOSmRNa,iNOSmRNA expression level in rat heart and lung were measured. RESULTS:The level of NO,ET-1 in the experimental groups were significantly higher and ADM was significantly lower than that of control group,and these changes can be restrain in injected L-NAME experimental groups.CONCLUSION:NO may play an important role in the change of ADM,ET-1 after acute hypoxia.
