摘要
目的研究念珠菌药敏试验的最佳条件,并测定5种抗真菌药物对78株念珠菌的敏感性。方法采用稀释法测定最小抑菌浓度,并计算50%菌株的最小抑菌浓度(MIC50)和90%菌株的最小抑菌浓度(MIC50)。结果药敏测定条件以10g/L蛋白胨和20g/L葡萄糖组成的沙保弱培养基、pN7.0和接种菌量为107~108CFU/ml为最好。两性霉素B、5-氟胞嘧啶、酮康唑和克霉唑MIC50分别为0.78,12.5,12.5,6.25μg/ml;MIC90分别为1.56,25,>25,25μg/ml。氟康唑MIC50为25μg/ml。除两性霉素B外,其余4种抗真菌药物对白色念珠菌的作用强于热带念珠菌。结论念珠菌药敏试验对选择有效抗真菌药物及研究耐药株的发生机制具有现实意义。
The best conditions of susceptibility testing of the Candida spp.were studied and five antimycotics were tested for susceptibility against 78 Candida spp.Methods MIC,including MIC50 and NIC90,was determined with broth dilution method.Results The best results of susceptibility testing were found in the Sabouraud media with 10 g/L peptone,20 g/L glucose,PH 7.0 and the inoculum concentration of 107108 CFU/ml.The MIC50 of amphotericin B,5-fluorocytosine,nizoral and kemazole were 0.78,12.5,12.5,6.25μg/ml,respectively,the MIC90 of these drugs were 1.56,25>25,25μg/ml,respectively.The MIC50 of fluconazole was 25μg/ml. Besides amphotericin B,other four drugs had better activity against Candida albicans than Candida tropicalis.Conclusion The susceptibility testing of the Candida spp.has practical significance in selecting the effective antimycotics and studying the developing mechanism of resistance isolates.
