枸杞多糖对人肝癌细胞HepG2生长的影响及其分子机制 被引量:13

摘要 目的探讨枸杞多糖(Lycium barbarumpolysaccharides,LBP)对人肝癌细胞HepG2生长的影响及可能机制。方法人肝癌HepG2细胞用100-1 000 mg/L LBP处理1-5 d,分别用MTT方法、流式细胞术检测细胞增殖、细胞周期和细胞凋亡。用Western blotting分析周期蛋白(cyclin)和周期蛋白依赖性激酶(cyclin-dependentkinase,CDK)表达的变化。结果 LBP以质量浓度依赖方式抑制肝癌细胞的生长,阻滞细胞在G0/G1期,并诱导细胞凋亡;其分子机制为引起cyclin Dc、yclin E和CDK2蛋白表达下降。结论 LBP抑制人肝癌细胞HepG2增殖的机制包括阻滞细胞周期和诱导细胞凋亡。
