摘要
目的研究人类白介素(human interleukin,hIL)-10基因修饰的骨髓间充质干细胞(mesenchymal stem cell,MSC)对大鼠原位异种肝移植排斥反应的影响及机制。方法建立豚鼠(供体)对Wistar大鼠(受体)的非协调性异种原位肝脏移植模型。供受体各90只,随机分为三组:空白对照组(A组),MSC组(B组),hIL-10修饰MSC(hIL-10-MSC)组(C组),每组各30只。观察受体鼠存活时间、肝功能的变化,用逆转录聚合酶链反应(reverse transcription polymerase chain re-action,RT-PCR)、酶联免疫吸附试验(enzyme-linked immune absorbent assay,ELISA)检测移植肝的白介素(interleukin,IL)-12、IL-15、IL-18、IL-4、肿瘤坏死因子(tumor necrosis factor,TNF)-α及IL-10的mRNA表达情况及各细胞因子的含量。结果与A组和B组相比,C组大鼠的生存时间延长、肝功能好转,细胞因子IL-12、IL-15、IL-18、TNF-α含量及其mRNA表达明显降低,IL-4和IL-10显著升高(P<0.05)。结论 hIL-10-MSC对移植肝的保护作用可能与其抑制IL-12、IL-15、IL-18、TNF-α细胞因子的表达,促进细胞因子IL-4的表达有关。
Objective To study the effect and mechanism of human interleukin-10 modified mesenchymal stem cell(hIL-10-MSC)in the rejection of the heterologous orthotopic liver transplantation from guinea pig to rat.Methods Heterologous orthotopic liver transplantations were performed from guinea pig to rat.Ninety guinea pigs and ninety Wistar rats were selected as donors and recipients respectively,and they were divided into control group(Group A),mesenchymal stem cell(MSC)group(Group B)and hIL-10-MSC group(Group C).Survival time and liver function of recipients were observed.The mRNA level expression and the content of interleukin(IL)-12,IL-15,IL-18,IL-4,tumor necrosis factor(TNF)-α and IL-10 were respectively measured by reverse transcription polymerase chain reaction(RT-PCR)and enzyme-linked immune absorbent assay(ELISA).Results Compared with group A and B,the recipients in group C had prolonged survival time and better liver function.The content and mRNA level expression of IL-12,IL-15,IL-18,TNF-α were significantly decreased,while the level of IL-4 and IL-10 were significantly increased in group C(P0.05).Conclusion The immunosuppression effect of hIL-10-MSC on transplanted livers may result from the decreased expression of IL-12,IL-15,IL-18,TNF-α and increased expression of IL-4.
