摘要
目的:通过建立体外模拟CO2气腹环境,研究CO2气腹对人肾癌786-O细胞增殖的影响。方法:建立体外模拟CO2气腹环境,以14mmHg气压对人肾癌786-O细胞作用不同时间(2、4h),以培养于常规条件下肾癌细胞为对照,用MTT法检测细胞增殖情况;流式细胞仪检测细胞周期分布和细胞凋亡率;免疫细胞化学法检测增殖细胞核抗原(PCNA)和Survivin蛋白表达水平。结果:与对照组比较,CO2气腹处理后的两组人肾癌786-O细胞增殖速度明显加快(P<0.05),且48h后细胞增殖速度的加快与气腹作用时间呈直线相关(R2=1,P<0.01);细胞周期中处于增殖期的细胞比例显著增高(P<0.01);PCNA表达明显增强(P<0.01);细胞凋亡率亦显著减少(P<0.05),且凋亡率的减少与气腹作用时间呈直线相关(R2=1,P<0.01);Survivin表达显著增强(P<0.01)。结论:体外模拟CO2气腹环境,可促进人肾癌786-O细胞增殖,且随着接触CO2时间的延长,肾癌786-O细胞增殖速度加快。
Objective:Tostudythe effects ofsimulated carbon dioxide pneumoperiton eumenvironment on cell proliferation of human renal carcinoma 786-O cells in vitro.Methods:The renal carcinoma 786-O cells were exposed to simulated CO2 pneumoperitoneum environment for different time periods(2 h or 4 h)under the pressure of 14 mmHg and those cultured in normal experimental condition as control,then proliferation viability of 786-O cells was observed by MTT assay close behind the exposure;cell cycle and apoptotic rate were detected by flow cytometry,the protein expression of PCNA and Survivin were examined by immunocytochemistry 48 h after exposure respectively.Results:Compared with the control group,Both of the treated groups showed cell proliferation was significantly increased (P0.05),and the increasing had a linear relationship with exposure time(R2=1,P0.01);The proliferative index(PI)showed significant difference from that of control group(P0.01);In cell cycle,the percentages of S-phase and G2/M phase were significantly higher in treated groups than those in control group(P0.05);PCNA protein was found to be significantly upregulated in treated groups (P0.05).The apoptotic rates were significantlydecreased(P0.01),andthe decreasinghadalinearrelationshipwithexposure time;the expressions of Survivin,which related with apoptosis,were significantly increased(P0.01).Conclusion:Carbon dioxide could promote the proliferation of human renal carcinoma 786-O cells,and the proliferation would be sped up with the prolongation of exposure time.
