The determination of lithium in biomaterials by flame atomic emission spectrophotometry with micro-plate pulse sampling

摘要 目的:建立微孔板脉冲进样-火焰原子发射光谱法测定微量生物材料中痕量锂的方法。方法:移取50μl样品于96微孔板中,加50μl 1%硝酸混匀后直接喷入原子吸收仪,记录脉冲发射强度,标准曲线法定量。结果:优化的测定条件为:进样量100μl,乙炔流量1.2 L/m in,燃烧器高度6 mm,测定时间6 s,脉冲发射峰高定量。锂含量在0-40μg/L范围内线性良好,回归曲线方程为y=2.502x+0.349,相关系数r=0.9950,方法检出限0.4μg/L,相对标准偏差(RSD)为2.2%-3.1%,血清、尿样和唾液的回收率在86.4%-103.4%之间。结论:微孔板脉冲进样-火焰原子发射光谱法测定微量生物材料中的锂,该方法取样量少,操作简单快速,准确度和精密度良好,适用于微量生物材料中的痕量锂测定。 Objective:To establish a method for the determination of Lithium in micro biomaterial sample by FAES combined with pulse sampling.Methods: 50 μl samples were pipetted onto the 96-hole plate,mixed with 50 μl 1% HNO3 and sprayed into atom absorption spectrophotometry,quantified by pulse emission intensity.Results: Assay conditions were optimized as followed: The spraying volume was 100 μl,the flow of acetylene was 1.2 L/min,and the height of burner was 6 mm.Lithium yielded good linear relationship between 0~40 μg/L with regression equation y=2.502x+0.349 and correlation coefficient r=0.9950.The relative standard deviation,recovery and the detection limit of the proposed method were 2.22%~14.8%,86.4%~103.4% and 0.4 μg/L respectively.Conclusion: This proposed method was easy,fast,accurate and suitable for the determination of Lithium in micro biomaterial sample with minimal 100 μl sample.
