摘要
采用氯胺丁改良法和过碘酸钠氧化法,进行抗AFP多抗或单抗直接双标记131Ⅰ和MMC,过Sephadex-G50柱并以r计数仪与紫外分光光度仪同步监测分离、收集标记物。经测定多抗和单抗双标记物,标记比分别为70.3MBq∶mg∶2.2μg和59.2MBq∶mg∶1.8μg;放化纯度98%和97%;131Ⅰ利用率83%和88%;抗体回收率62%和74%;标记后与标记前相比抗体活性均无明显降低。置4℃5d和10d时,抗体活性不受影响;放化纯度分别降为95%和92%、91%和88%。经裸鼠人肝癌模型放免显像,72h即见肿瘤区有放射性浓集,至120h肿瘤显像最清晰。结果表明,多抗和单抗的双标记物的生物、理化性质稳定。为免疫导向治疗肝癌提供了一种新型的“双弹头”导向药物。
Anti AFP polyclonal antibody(PcAb) or monoclonal antibody(McAb) was labeled
directly with 131 Ⅰ and Mitomycine C(MMC) by modified Chloramin T method and sodium
periodate oxidation method respectively The double labels were gained when the mixture
passed through Sephadex G 50 column,which were monitored,separated synchronously with
Gamma counter and ultraviolet spectrophotrometry The label ratioes of the labeled PcAb and
McAb were( 131 Ⅰ∶anti AFP PcAb or McAb∶MMC)70 3MBq∶mg∶2 2μg and 59
2MBq∶mg∶1 8μg respectively The radiochemical purity,the rates of 131 Ⅰ utilization
and antibody recovery were 98%,83%,62% and 97%,88%,74% respectively There was no loss of
immunoreactivity before and after label In vitro,the immunoreactivity was not affected when the
label antibody was stored at4℃ on day 5 and day 10 The radioimmunoimage in nude mice
bearing liver cancer model showed that radioactive accumulation was seen in the tumor area on
the day 3,and the tumor image was most distinct on day 5 These results indicate that the
biological and physicochemical properties of the labeled antibody is stable It has provided a
new type“dual warhead”targetting drug for RIT in the treatment of patients with PLC
