摘要
目的观察商陆水煎剂能否减轻阿霉素肾病大鼠肾小球细胞外基质(extracellular matrix,ECM)成分蓄积。方法单侧肾脏切除并重复尾静脉注射阿霉素建立肾病大鼠模型,随机分为模型组、商陆组、依那普利组,对照组为假手术大鼠。给药14wk,肾脏石蜡切片PASM染色测定肾小球基质相对面积;免疫组化方法检测层粘连蛋白(laminin,LN)与纤连蛋白(fibronectin,FN)及基质金属蛋白酶-2(matrix metalloproteinases-2,MMP-2)表达。结果①商陆组与依那普利组ECM相对面积均显著低于模型组(P<0.05),二者间无统计差异;②与模型组比较,商陆组与依那普利组肾小球LN、FN表达下调(P<0.05),MMP-2表达上调(P<0.05)。结论商陆水煎剂对减少阿霉素肾病大鼠肾小球ECM蓄积、延缓肾病进展有一定作用。
Objective To observe the effect of pokeberry root decoction(PRD) on the glomerular extracellular matrix(ECM) in rats of doxorubicin nephrosis.Methods Fifty Sprague-Dawley rats were randomly divided into four groups: control group,model grou,PRD group and enalapril group.Doxorubicin nephrosis model was induced with unilateral nephrectomy and repeated vena caudalis injection of doxorubicin.After 14 weeks,paraffin sections of the renal tissue were stained by PASM and 1immunohistochemistry methods.The changes of ECM,fibronection(FN),laminin(LN),and matrix metalloproteinase-2(MMP-2) were quantificationally analyzed by computer image analysis.Results ①Relative areas of the glomerular ECM in PRD group and enalapril group were all decreased as compared with model group(P 0.05)② Compared to model group,the expressions of LN and FN were decreased,while MMP-2 was increased in PRD and enalapril groups(P 0.05).Conclusions PRD can partly decrease the glomerular ECM accumulation,which is useful for delaying the progression of nephrosis induced by doxorubicin.
