D-异苯环戊胺对营养性肥胖模型大鼠的减肥作用研究 被引量:5

Anti-obesity Effect of D-iso-phenylcyclopentylamine on Diet-induced Obese Rats

摘要 目的:探讨D-异苯环戊胺的减肥作用。方法:建立营养型肥胖大鼠模型,选取造模成功的50只♂大鼠分为5组,即模型组、西布曲明组及D-异苯环戊胺大、中、小剂量组,另设正常对照组(10只),共6组。各实验组分别连续灌胃给予相应药物4周,于末次给药并禁食12 h后,测量体长、尾长计算Lee’s指数,并观测各组大鼠体质量、肾周脂肪重量、脂肪系数、睾周脂肪细胞数量、脂肪细胞大小以及血脂、血糖水平的变化情况。结果:与模型组比较,D-异苯环戊胺各剂量组大鼠体质量、Lee’s指数、肾周脂肪质量、脂肪系数及睾丸周围脂肪细胞大小、血糖、血脂水平均明显降低(P<0.05或P<0.01),并存在一定的量效关系;其中D-异苯环戊胺大剂量组与西布曲明组各指标比较无显著性差异(P>0.05),中、小剂量对大鼠体脂和脂肪细胞的影响不及西布曲明(P<0.05或P<0.01)。结论:D-异苯环戊胺有较明显的减肥作用。 OBJECTIVE: To investigate the anti-obesity effects of D-iso-phenylcyclopentylamine(D-IPCPA) on diet-induced obese rats.METHODS: Diet-obese models were induced successfully,50 male model rats were randomly divided into model group,sibutramine group,and D-IPCPA groups(high-dose,middle-dose and low-dose).Another 10 rats were involved in normal control group.Those groups were given sibutramine and D-IPCPA via i.g.gtt for 4 weeks.Those groups were fasting 12 hours after the last administration.Then the length of body and tail were measured to calculate Lee's indexes.Several indexes were detected in respect of body weight,perirenal fat weight,fat index(the ratio of fat weight to body weight),the number and size of adipocyte around testis,the level of blood lipid and blood glucose.RESULTS: Compared with model group,the body weight,Lee's index,perirenal fat weight,fat index,the size of adipocyte around testis and the levels of blood lipid and blood glucose in D-IPCPA groups(high-dose,middle-dose and low-dose groups) were significantly decreased(P〈0.05 or P〈0.01).Certain dose-effect relationship was observed in D-IPCPA groups.There was no significant difference in above indexes between D-IPCPA high-dose group and sibutramine group(P〉0.05).The effects of middle-dose and low-dose of D-IPCPA were inferior to sibutramine(P〈0.05 or P〈0.01).CONCLUSION: D-IPCPA has an obvious anti-obesity effect.
