摘要
目的:探讨干扰素(IFN)对肝癌细胞HLA-DR表达的调节作用。方法:采用免疫组化ABC法和ELISA法分别检测了不同剂量的IFN-γ或IFN-α刺激前后HLA-DR在HHCC、SMMC-7721、BEL-7402和HCC-9204等4种人肝细胞癌细胞系以及正常人肝细胞系QZG的表达情况。结果:IFN-γ或IFN-α刺激之前,5种细胞系中只有少数的SMMC-7721细胞可表达HLA-DR;刺激之后,对HLA-DR的表达5种细胞系都有所增强,并且与IFN-γ的剂量呈正相关,QZG不如肝癌细胞明显,IFN-γ的作用强于IFN-α。结论:IFN可以上调肝癌细胞对HLA-DR的表达。关键词:##4干扰素;;HLA-DR;;肝细胞癌;;免疫组织化学;;
Object: To require into the regulative effect of interferon (IFN) on HLA-DR expression in human heaptocellular carcinoma (HCC). Methods: The expression of HLA-DR in 4 HCC cell lines, HHCC, SMMC-7721, BEL-7402,HCC-9204 and a normal liver cell line QZG was detected with immunohistochemical ABC and ELISA methods before and after stimulated by different doses of IFN-γ or IFN-α. ResultS: Only a little part of SMMC-7721 cells expressed HLA-DR before stimulation by IFN-γ or IFN-α, and the others were HLA-DR negative; HLA-DR expression enhanced in all of the 5 cell lines after stimulation, and was correlated with the dose of IFN, QZG was less obvious than HCC cells, the effect of IFN-γ was obvious than IFN-α. Conclusion: IFN can induce HLA-DR expression in HCC cells and make self-immunological adjustment in HCC on some extent.
