摘要
应用抗癌药阿霉素成功地诱导培养的人肝癌细胞株HCC-9204肝癌细胞发生凋亡。用20μmol·L^(-1)阿霉素作用肝癌细胞3h,换培养液继续培养12h后,肝癌细胞DNA结构发生断裂,电泳呈梯状。肝癌细胞形态改变出现细胞膜小泡和凋亡小体形成等凋亡细胞特征。经透射电镜和扫描电镜观察显示,在体积缩小胞膜皱缩的肝癌细胞表面产生一种球形小体,有的肝癌细胞崩解为碎片状。形态学观察结构提示,肝癌细胞产生的球形小体可能是一种与细胞凋亡有关的功能性结构。细胞崩解可能是肝癌凋亡细胞灭亡的形式之一,可能与细胞骨架破坏有关。
The apoptosis of human cultured hepatocellular carcinoma cells was induced by adriamycin,anticancer drug, in HCC-9204 cell line. The apoptosis-associated DNA fragmentation occured , referred to as a"DNA ladder", in hepatoma cells treated with adriamycin for 3 h at 20 μmol·L ̄(-1) and post-incubated in drug-free medium for 12 h. The characteristics of apoptosis, such as the formation of cell-membrane blebs and apoptotic bodies, were observed morphologically in hepatocellular carcinoma cells. In transmission and scanning electron microscopy, it has been shown that one kind of balllike body produced on the surface of hepatoma cell with volume reduction and shrinkage cell-membrane was demonstrated,and also the explosion of some hepatoma cells into fragmentation was found. The morphological changes of hepatoma cells imply that the ball-like body yielded by hepatoma cells is one kind of functional structure related to apoptosis,probably. Perhaps,the explosion of hepatoma cells is one kind of mode of apoptotic cells for disappearing,which is maybe related to the collapse of cell-skeleton.
