摘要
本文应用热启动聚合酶链反应技术,快速扩增出特异的水蛭肽基因,其DNA为221 bp,与理论预计相符。同时研究了不同退火温度对扩增反应结果的影响,确定了退火温度为50℃时的扩增反应结果较好。为从基因水平上进一步探讨水蛭肽基因克隆、表达与调控规律奠定了基础。
The gene of the peptide with anticoagulation activity in leech was amplified specifically by using preamplification heating PCR. The gene's molecular weight is in accordance with predict, with 221 bp. The effects of different annealing temperature were observed. 50℃ was better annealing temperature. Further research on cloning and expressing of the gene is being carried on.
