摘要
采用ISSR标记对中国北方地区的8个山荆子居群进行了遗传多样性分析,以探讨山荆子在苹果属植物演化过程中的作用.结果显示:(1)从55个ISSR引物中,筛选出11个多态性引物,共扩增出71个位点,其中多态性位点69个,多态位点达97.47%.(2)在物种水平上,有效等位基因数Ne=1.4670,Neis基因多样性(H)为0.2846,Shannon信息指数(I)为0.4399,多态位点百分率(PPB)=80.22;基因流(Nm)为1.6591,居群间基因分化系数(Gst)为0.2316,Φst值为0.2360.(3)聚类分析结果显示,在遗传距离为0.1555、0.1369、0.1258处8个山荆子居群分别聚为两大类、三亚类、四小类,但居群聚类位置与地理位置无明显的相关性.(4)山荆子的居群内遗传多样性贡献率占76.40%,而居群间的贡献率占23.60%,说明山荆子的遗传多样性主要分布在居群内,但居群间的遗传分化程度也较高.由此推测,山荆子的起源中心在华北和东北范围内,其中灵空山(山西)和塞罕坝(河北)居群可近似为核心居群;ISSR标记与花部的一些性状可能有连锁关系.
In order to shed light on the role of Malus baccata about the genus Malus evolution we analyzed the genetic diversities of 8 north populations of M.baccata by ISSR markers.The results showed that:(1)71 bands were amplified with eleven polymorphic primers screened from 55 random primers,polymorphic loci of which were 69,and their polymorphism percentage was 97.47%.(2)At the species level,effective alleles were 1.618,Nei’s genediver sity (H) was 0.284 6,Shannon’s information index (I) was 0.439 9,the percentage of polymorphic loci is 80.22%.Gene flow among populations (Nm) was 1.659 1.The coefficient of gene differentiation was Gst=0.260,Φst=0.236 0.(3)Based on Nei’s genediver,cluster analysis by UPGMA show that all populations could be divided into two large groups,three subgroups,four small groups,respectively in Nei’s genetic distance 0.155 5,0.136 9,0.125 8,respectively.However,clustering and geographic location was no significant correlation.(4)It was 76.40% of genetic diversity within populations,and was 23.60% of genetic diversity among populations.The genetic diversity within populations were mainly distributed,the genetic differentiation among populations were also high.Based on these results,the origin center of M.baccata was surmised in the context of Northeast and North China,Mt.Lingkong and Saihanba populations were like to core populations,in addition,there were possible genetic linkage between ISSR markers and some flower characters.
