从八角茴香中提取莽草酸工艺研究 被引量:4

摘要 [目的]研究八角茴香中莽草酸提取和纯化工艺。[方法]以八角茴香为原料,水蒸气蒸馏,回收八角茴香油,对残渣用水回流提取,提取液用711树脂富集,粗提物搅拌沸石粉,柱层析,乙酸乙酯洗脱,重结晶得到莽草酸。[结果]利用711树脂分离莽草酸最佳工艺条件为:上柱液中莽草酸浓度5.876 mg/ml,流速1.0 BV/h,动态交换吸附率达96.8%,洗脱剂为pH值3的浓度70%乙醇溶液,洗脱流速1.0 BV/h,解吸率达94.5%。莽草酸粗提物与沸石粉的质量比为1∶10,搅拌,柱层析,乙酸乙酯洗脱,洗脱率达到96.9%,真空浓缩至干,甲醇-水混合结晶,得到莽草酸含量为98.16%的提取物,收率为8.86%。[结论]该工艺成本低,收率高,适宜于莽草酸的提取。 [Objective]The research aimed to study the extraction and purification process of shikimic acid from Illicium verum Hook f.[Method] Illicium verum Hook f was used as raw materials to carry out steam distillation,recycling star anise oil,on the residue of water extraction and extract enriched with 711 resin,crude extracts mixing zeolite powder,column chromatography,ethyl acetate elution,recrystallization.And shikimic acid was got.[Result] The optimum conditions of the separation of shikimic acid with 711 resin on the column was as follows:the concentration of shikimic acid solution 5.876 mg/ml,velocity 1.0 BV/h,dynamic exchange adsorption rate 96.8%,eluting agent for the 70% ethanol solution of pH 3,elution flow rate 1.0 BV/h.And elution yield reached 94.5%.With shikimic acid extracts and the zeolite powder mass ratio of 1∶10 for mixing,column chromatography,ethyl acetate elution rate reached 96.9%.With vacuum concentration to dryness,methanol-water mixed crystal,shikimic acid content reached 98.16% of the extract,and the yield was 8.86%.[Conclusion] The process had the advantage of low cost,high yield,which was suitable for the extraction of shikimic acid.
