摘要
以新鲜猪肺为材料,Sepharose4B偶联胰蛋白酶作为亲和层析介质,从猪肺中分离纯化高比活猪肺抑肽酶。结果表明,猪肺的抑肽酶含量为1.86mg/kg,酶比活力达4593U/mg;猪肺抑肽酶耐热性好、对pH敏感性低,在280nm处有最大光吸收,对胰蛋白酶有很强的抑制作用。
The aprotintin with high activity was extracted and purified by affinity chromatography, based on crosslinking sepharose4B immobilizing trypsin.The results showed that the aprotinin yield was 1.86 mg/kg,the special activity of aprotinin reached 4 593 U/mg.The aprotinin was not only high heat-resistance,but also low pH sensitivity. And it had light absorption at 280 nm and strong inhibition to trypsin.
