应用高速逆流色谱法分离茎点霉属真菌代谢物中脂肪酸成份的研究 被引量:3

摘要 利用高速逆流色谱对一株茎点霉属内生真菌的代谢物质脂肪酸部分进行了分离。采用的两相溶剂系统为正庚烷:乙酸乙酯:甲醇:水=8.5:1:8.5:1,上相做固定相,下相做流动相。分离得到三个脂肪酸组分A5、A6、A7,经高效液相分析,纯度都在99%以上,其回收率分别为96.3%、93.5%,94.8%。经EI-MS、1H-NMR测定,确定A5为硬脂酸,A6为油酸,A7为亚油酸。 High speed counter-current chromatography (HSCC) was applied to the separation of fatty acids from Phoma endophytic fungi. A biphasic solvent system composed of n-heptane : ethyl acetate : methanol : water=8.5 : 1 : 8.5 : 1 was used. The top phase was chosen as the stationary phase, and the bottom phase as the mobile phase. The fatty acid components A5, A6, A7 were collected by HSCCC separation. Through HPLC analysis, the purities of these fatty acids were higher than 99 percent with recoveries of above 93.5 percent. The chemical structures of A5. A6. A7 were identified by EI-MS, ^1H-NMR as stearic acid. oleic acid and linoleic acid.
