摘要
作物品种纯度鉴定是当前国内外种子检验的一大难题。本文以小麦为材料,利用SDSPAGE电泳技术,分析了10个品种种子的醇溶蛋白图谱。结果表明,同一品种不同贮藏年限种子的醇溶蛋白组分没有发生变化,电泳图谱基本一致;而不同品种种子的醇溶蛋白条带差异明显,B区和C区的主带数目是品种的“指纹”特征,可用于品种鉴定。本文还对几项电泳条件进行了讨论。
The identification of cultivar purity is one of the most difficult problems in seed testing in the world nowadays,The banding patterns of gliadin from single seeds of 10 wheat cultivars by SDS—PAGE technology were obtained in the experiment,The result showed that the components of gliadin didn't change in seeds of different storage periods of the same cultivar, and their electrophoregram was similar. But the banding patterns of gliadin from single seeds differed obviously among wheat cultivars,and as a'fingerprint' character of wheat cultivars, the number of major bands in zones B and C could be used for identification of cultivars. Several conditions of electrophoresis were also discussed in the paper.
