摘要
目的研究枇杷叶紫珠中高极性化学成分,为其药效成分及作用机制的深入研究奠定基础。方法采用甲醇提取,不同极性溶剂分布萃取,获得正丁醇萃取部位;采用大孔树脂、反相硅胶、Sephadex LH-20、Toy-opearl HW-40等色谱技术进行分离纯化,综合运用紫外光谱(UV)、红外光谱(IR)、质谱(MS)及核磁共振光谱(1H NMR、13C NMR、1H-1H COSY、HSQC、HMBC)等现代波谱学方法和技术进行化合物的结构鉴定。结果自正丁醇萃取部位中共分离得到8个化合物,鉴定出其中6个化合物的结构,分别为毛蕊花糖苷acteoside(1),连翘苷forsythosides B(2),肉苁蓉苷C cistanoside C(3),异毛蕊花糖苷isoacteoside(4),肉苁蓉苷D cistanosideD(5)和独一味苷A lamiophlomioside A(6)。结论所鉴定的6个化合物均为首次自该种植物中分离得到,其中肉苁蓉苷C(3)、肉苁蓉苷D(5)和独一味苷A(6)为首次自紫珠属中分离得到。苯乙醇苷类成分为枇杷叶紫珠中高极性部位主要化学成分。
Objective To study the high-polarity chemical constituents of Callicarpa kochiana Makino,thus to supply evidence for its pharmacological research and therapeutic mechanism.Methods The chemical constituents of Calli-carpa kochiana Makino were extracted by methanol and different polar solvents to obtain n-butanol extract.Then n-butanol extract was isolated and purified by macroporous resin,reversed phase silica gel,Sephadex LH-20 and Toyopearl HW-40.The structure of n-butanol extract was identified by spectral analysis of ultraviolet spectrum,in-frared spectrum,mass spectrum,and NMR spectrum.Results We obtained 8 chemical compounds from n-butanol extract.Of them,six phenylethanoid glycosides were identified as acteoside(1),forsythosides B(2),cistanoside C(3),isoacteoside(4),cistanoside D(5),lamiophlomioside A(6).Conclusion Compound 3,5 and 6 is isolated from Callicarpa genus plant for the first time,and phenylethanoid glycosides are the main chemical constituents from n-butanol fraction of Callicarpa kochiana Makino.
