不同浓度苦参碱对肝癌细胞增殖及JAK-STAT信号通路的影响 被引量:13

摘要 目的:观察不同浓度苦参碱对肝癌细胞SMMC-7721增殖和JAK-STAT通路的影响。方法:苦参碱作用于肝癌SMMC-7721细胞,用MTT法检测不同浓度苦参碱对SMMC-7721细胞增殖的抑制作用;RT-PCR法检测不同浓度苦参碱对SMMC-7721细胞stat3、stat5、c-jun mRNA的影响,Western blot法检测不同浓度苦参碱对肝癌SMMC-7721细胞STAT3、STAT5、P-STAT3、P-STAT5、C-JUN蛋白表达的影响。结果:苦参碱能够抑制SMMC-7721细胞增殖,呈时间-剂量依赖性。苦参碱能下调stat3、stat5、c-jun mRNA表达水平,降低STAT3、STAT5、P-STAT3、P-STAT5、C-JUN蛋白表达量,有剂量依赖性。结论:苦参碱不仅能直接杀伤肿瘤细胞,而且能抑制JAK-STAT信号通路,从而抑制肿瘤细胞的增殖。 Objective:To observe the effect of matrine on cell proliferation and JAK-STAT signaling transduction pathway in SMMC-7721 cells.Methods:Treated with different concentration matrine,the inhibitory on proliferation was detected by MTT,the mRNA expression of stat3,stat5,c-jun in SMMC-7721 cells was assessed with RT-PCR. Western blot detected the protein expression of STAT3,STAT5,P-STAT3,P-STAT5,C-JUN in SMMC-7721 cells. Results:Matrine significantly inhibited SMMC-7721 cells proliferation in time and dose-dependent manner. Matrine could degrade the expressional levels of stat3,stat5,c-jun mRNA and STAT3,STAT5,P-STAT3,P-STAT5,C-JUN protein in dose-dependent manner. After treat with matrine,the fluorescence intensity of STAT3,STAT5,P-STAT3,C-JUN was significantly weaker than that in control group,The high dose group was more obviously. Conclusion:Matrine could significantly down-regulate the mRNA expressional evels of stat3,stat5 in SMMC-7721 cells and decrease the phosphorylation of STAT3 and STAT5 protein and block their downstream signal C-JUN,thus it could inhibit the proliferation of SMMC-7721 cells.
