摘要
目的 :探讨苦参素注射液对肿瘤血管形成的抑制作用。方法 :通过建立动物肿瘤模型 ,运用分别实验治疗 ,结合免疫组化 ,检测其抑瘤率和肿瘤内血管密度 ,以及瘤体血管内皮生长因子 (VEGF)和碱性成纤维细胞生长因子 (bFGF)表达的改变。结果 :苦参素具有明显的抑制S180 肉瘤作用 ,其大剂量组抑瘤率为 31.36 %。中、大剂量苦参素组的S180 瘤体内微血管密度均明显低于对照组 ;免疫组化显示苦参素大剂量组可抑制S180 瘤体内VEGF、bFGF的表达。结论 :苦参素对小鼠S180 肉瘤有一定的抑制作用 ,对S180 肉瘤血管形成有明显的抑制作用 ,降低VEGF ,bFGF的表达可能是其抑制肿瘤血管形成的主要机制之一。
Objective: To investigate the inhibiting effect of oxymatrine on mice's angiogenesis from S 180 sarcoma. Method: The tumor inhibitory rate and the vascular density were tested in animal tumor model by experimental treatment. The expression of VEGF and bFGF were measured by immunohistochemistry.Result: When the high dose was used, the tumor inhibitory rate of the sophora flarescens ait was 31.36%; the vascular density of S 180 sarcoma was lower than the control group;and the expression of VEGF bFGF was down-regulated when the high and middle dose were used.Conclusion: Oxymatrine has inhibitory effect on S 180 sarcoma and inhibitory effects on angiogenesis. Its mechanism may be assiociated with the down-regulating of VEGF and bFGF expression.
